Sulforaphane is a substance present in various vegetables such as broccoli, it prevents and reduces the expression of long non-coding RNA fragments associated with cancer. So concludes a recent study by the University of Oregon, recently published in the Journal of Nutritional Biochemistry.
Consumption of certain vegetables such as broccoli reduces the risk of developing prostate cancer. Various studies suggest that this is due to a compound, sulforaphane, which is produced when these vegetables are chewed and which acts against cancer cells. However, the exact molecular mechanisms by which sulforaphane has a preventive effect on prostate cancer are not known in detail.
A recent study from the University of Oregon has just discovered one of these mechanisms, finding that sulforaphane prevents and reduces the expression of long non-coding RNA fragments associated with cancer.
LncRNA or long non-coding RNA fragments are RNA fragments longer than 200 nucleotides or RNA units that do not encode any protein. The existence of these fragments in mammalian cells has long been known. Nevertheless, in recent years, their functional significance has begun to be revealed. LncRNAs can regulate gene expression and regulate protein activity and are involved in a variety of biological processes. In addition, their change can contribute to the development of pathological processes.
Molecular structure of sulfofafan
In their work, the researchers considered whether an interaction between lncRNA and diet could occur, and whether sulforaphane could exert its preventive effect via lncRNA.
To address this issue, the team analyzed the effect of sulforaphane on the expression of cancer-related lncRNAs in human prostate cells. While performing RNA sequencing, the researchers noticed that sulforaphane affects the expression of almost different lncRNAs, both in healthy cells and in prostate tumor cells. Further, sulforaphane treatment restored the expression of some of the altered lncRNAs expressed in cancer cells.
Among lncRNAs whose expression varies in the presence of sulforaphane, researchers emphasize the role of LINC01116. Levels of this lncRNA appear to be elevated in various types of cancer, such as glioblastoma, colon cancer, or prostate cancer, and its expression is strongly suppressed by sulforaphane. Furthermore, LINC01116 promotes tumor cell proliferation.
“We have shown that sulforaphane treatment could normalize the level of this lincRNA,” said Laura Beaver, a researcher at the Linus Pauling Institute and the College of Public Health and Human Sciences and the first author. “This could be more than cancer prevention. It would be of great value if we could develop methods to slow the progress of cancer by helping it to become invasive. ”
The research team points to the need for further studies regarding sulforaphane and the role of LINC01116. Of particular interest will be the determination of the targets on which lncRNA acts, which is still needed in more studies.
“The data support sulforaphane as a promising dietary anticancer agent and suggest that it could exert its chemopreventive effects through multiple mechanisms, including regulation of lncRNA expression,” the authors conclude. “Our data reinforces the idea that lncRNAs are an exciting new way to investigate chemoprevention and that dietary compounds can affect their expression.”
