Modern molecular biology enables rapid diagnosis of the presence of bacterial DNA with just one biological test. Nevertheless, the possible survival of the bacterium in the infected organism must arouse some prudence in the interpretation of these experiments.

Pathology in the urogenital tract is most likely limited to the mucous membrane that lines the cavities (pharynx, eyes, vagina, cervix, intestines, bladder) including also the internal glands that can represent an infectious reservoir or complicate mucosal infection.

It is considered that all serotypes from the group of trachomonas can be involved, but it is more often and more seriously about group L (L1, L2, L3), which is the biggest cause of venereal lymphogranuloma, and from groups D to K (D, E, F, G, H, I, J, K).

After infection, a person develops inflammation of the urethra, which can sometimes occur simultaneously with a gonorrheal infection that manifests itself quickly, which is why it can go unnoticed and without establishing a diagnosis. Unable to accumulate enough energy for independent development, chlamydia is forced to the most developed parasitic organisms such as eukaryotic cells that allow parasites to become energy-dependent parasites themselves, which morphologically leads to an intracellular colony with different structures inside the infectious vacuole called: inclusion. Only small dense and compact particles, called elementary corpuscles (0.2-0.4 mcm), are infectious. Material and method of research Chlamycin

–  Cell substrate:  the Mr Coy line (ATCC CRL-1696) from the Laboratory “Virceil” was used

–  Growth medium: DMEM (Dominique Dutscher SAS) with 10% fetal bovine serum (SFB) )Dominique Dutscher, 4.5 g/l glucose, 2mM L Glutamine, sodium pyruvate and 3.7 g/l Na H COOH, gentamicin 20 mcg/ml.

– Cell suspension:  cell membrane washed with EBSS without Ca Mg nor HC03Na-phenol, separated with trypsin in PBS concentration 0.25%. The effect of trypsin on PBS is blocked by EBSS with 10% SFB and regulated to 2×10*5 cells with a diameter of 14 mm after counting in a Malassez chamber.
the suspension is distributed in a tube “Sterilin” with a flat base with one round object for covering with a diameter of 14 mm, and the incubation of the tube is carried out in a heater with 5% CO2, for 24 hours.

–  Inoculum: lineage (family) L” (Vircell) of Chlamydia trachomatis was used, originating from an infected cell culture of 72h, whose particles were obtained after a series of freeze-thaws, and adjusted to a concentration of 10,000 cfi/ml. Add 100 µl to each tube and centrifuge horizontally at 2500 rpm for one hour at 35 degrees. After centrifugation, the tube is incubated at 37° for two hours.

Incubation sample: Aspirate the incubation sample and aliquot 1 ml of the growth sample to which one final concentration of 1 mcg/ml cycloheximide (Albaran 424454561 – Sigma-Aldrich-Quimica) is added-

–  CHLAMYCIN: the product under investigation appears as a dark green liquid and corresponds to a mixture of tinctures:

– T.uvea ursi folium, T.juniperis fructus, Hyperi herba, T.serpilli herba, T.caryophylli flos, T.plataginis lancelotae folium, in a 60% ethanol solution.

– The presence of ethanol in the preparation forced us to evaluate the action of incubation agents with a similar concentration of ethanol itself, according to the same solutions as the product in the incubation agent.

Before dissolution, the gross product was centrifuged for one hour at 4000 rpm in order to remove any residue (waste) that could damage the cell membrane. Four solutions of the product in the incubation medium were made: 1/10, 1/20, 1/40, 1/80. For each experiment, 4 tubes were used, thus creating 4 batches.

Reading:  During 48 hours of incubation at 36° in a medium of 5% CO2, the incubation medium is discarded, the cell membrane is washed with 1 ml of EBSS, the cell membrane is fixed with cold acetone (4°C) for 10 min. Add 100 mcl of anti-generic monoclonal antibody, mixed with fluorescein (30 min at 37°C). Microscopic observation of epi-fluorescence (Leitz, with filters BG 12, 535 K, eyepieces 10X, one of them with a micrometer line). Inclusions from the entire round blanket are counted.

RESULTS AND DISCUSSION

For the tubes that represented the means of incubation, the value of the average count is 733 inclusions (in order: 580,944,780, 620) per one tube.

In medium with ethanol alone (ethanol dissolved at 1/10, 1/20, 1/40, 1/80 in incubation medium) it shows a significant decrease at the lowest solution (1/10) with also a significant decrease in inclusion size.

This reduction in inclusion size is also observed in all solutions containing the active products, with inhibitory activity being most prominent in the highest solution with CHLAMYCIN.

If the activity of the product is compared with the possible activity of ethanol itself, inside the incubation medium, the idea that the global inhibitory activity is related to the presence of the solvent (ethanol) can be rejected, better than the activity itself, because it is not manifested in the lowest solution (1/10) any inhibitory effect compared to the activity of Chlamycin.

By analogy with the application of the minimum inhibitory concentration for bacteria, we consider a significant value< o = a 20% of the formed bodies of the inclusion.

It is clear that the tested product has inhibitory activity up to a 1/40 solution. It would be necessary to know whether in practical daily use, as a dietary supplement, these concentrations in the body can be achieved.

It would be interesting to have pharmacological data, knowing that the components of the CHLAMYCIN product are, as a general rule, powerful antioxidants (22, 23, 24) that act by stimulating the organic defense against attacks by various microorganisms. (25, 26)

Among them are uvae ursi folium and serpilli herba, which form an integral part of the product concentration. Conclusion

The properties of Chlamycin that we described previously, together with the direct mechanical aspect of individual ingredients (juniper: 27) promise a lot in helping to eradicate the infection and avoid possible and very common complications that are not clinically noticeable.

To these encouraging results, it is necessary to add a research study with the aim of adjusting the most favorable dosage for this type of infection.

    Do NOT take oregano oil diluted in water or directly on the skin. Oregano oil BURNS like crazy and MUST be diluted with a carrier OIL. Water won’t do the trick, because they don’t mix. You will burn your mouth, stomach and skin by following this article’s advice. Webmaster, please change this advice ASAP.

      My friend there is a difference between Oregano oil and essential Oregano oil. We don’t recommend the usage of essential Oregano oil or any other type of essential oil. Please for start make difference between this 2.
